Diet-Induced Obese Mouse Model
Study objective
Researchers investigated the effects of retatrutide in mice with diet-induced obesity and metabolic dysfunction-associated steatohepatitis (MASH), a preclinical model commonly used to study obesity and metabolic disease.
Methods
Male mice were fed a high-fat diet to induce obesity before receiving retatrutide treatment. Researchers monitored:
* Body weight
* Food intake
* Body composition
* Insulin resistance
* Liver fat accumulation
* Blood lipid concentrations
* Energy expenditure
Results
Compared with untreated controls, researchers observed:
* 31% reduction in body weight during the treatment period.
* Significant reductions in fat mass, with some reduction in lean mass.
* Marked decreases in food intake during the initial days of treatment.
* Significant improvements in insulin resistance (measured using HOMA-IR).
* Reduced liver fat accumulation and hepatic steatosis.
* Lower liver triglyceride and cholesterol content.
Interestingly, in this particular study, energy expenditure was not significantly increased, suggesting that the observed weight loss in this model was driven primarily by reduced food intake rather than a measurable rise in metabolic rate.

The same investigators also evaluated retatrutide in obese hamsters maintained on a high-fat, high-cholesterol dietary model.
